Van der Sar should be back on Wednesday after he missed Saturday's draw against Blackburn because of a groin strain.
"I’d love to get my hands on it again. It’s great to win the Champions League once, and a lot of people will do that, but to win it twice is a big incentive," Van der Sar said.
Wayne Rooney should also feature at the Nou Camp despite suffering a hip in jury against Blackburn.
Owen Hargreaves and Anderson are also in contention after they were ruled out of the game at Ewood Park with minor injuries.
